In fact, I don't think it was ever established why the show was titled "Elfen Lied".
Unless I'm A.D.D. and I didn't catch when they said it.

They call it Elfen Lied because it's German for Elf Song.
In the manga, one of Yuka's friends tought Nyu how to sing the song.
So they named the anime/manga after that song.
